Abstract
Abstract
This study evaluated the antidiabetic activity and safety of Bululawang sugarcane (Saccharum officinarum) syrup (BSS) using in vivo and in silico approaches. Antidiabetic activity was assessed in alloxan-induced diabetic mice and an α-amylase inhibition assay, while safety evaluation included physiological observation, kidney function parameters, and blood urea nitrogen (BUN) analysis. Liquid chromatography–mass spectrometry (LC–MS) was used to identify bioactive compounds, followed by molecular docking to investigate their interaction with α-amylase. The results showed a trend toward reduced blood glucose levels at lower doses (10 mg/kg BW) during the 14-day treatment period. Toxicity evaluation revealed no adverse effects on body weight, kidney weight ratio, creatinine, urea, or BUN, which remained within normal physiological ranges. In the α-amylase inhibition assay, BSS exhibited inhibitory activity ranging from 49.139 to 81.798%, with the highest inhibition observed at 100 µg/mL. LC–MS analysis identified several bioactive compounds, including flavonoid glycosides and phenolic compounds. Molecular docking demonstrated that selected compounds exhibited binding affinities toward α-amylase, with 10-desmethoxystreptonigrin showing the strongest interaction (−8.0 kcal/mol), compared to acarbose (−7.7 kcal/mol). These findings suggest potential inhibitory activity against carbohydrate-digesting enzymes.
