Driven by urbanization and industrialization, land use in the middle and lower Yangtze River Basin has changed drastically and disturbed regional ecosystem services. Taking seven provincial administrative regions along the river as study areas, this paper quantified 2000–2020 ecosystem service value (ESV) via equivalent factor method and analyzed its spatial features and driving forces. Construction land surged over 90% with continuous cultivated land shrinkage; forest and farmland contributed over 75% to total ESV, while water bodies buffered ESV decline. ESV's Moran's I fell from 0.4982 (P<0.01) in 2000 toward random distribution, broken by intensive human activities. Economic expansion restrained ESV improvement, and natural-socioeconomic factors jointly dominated ESV differentiation. Cross-regional ecological governance is recommended to boost ecosystem sustainability.
